The Titan Krios G4 with Cold-FEG and Falcon 4i detector:

Krios #2 is a Titan Krios G4 300 kV high-end cryo-transmission electron microscope (cryo-TEM) designed for automated data acquisition.

Key Features and Capabilities of Krios #2:

  • E-CFEG: Equipped with a Cold Field Emission gun with a narrow energy spread that results in higher contrast images.
  • Automation: Includes a 12-sample autoloader and software for automated Single Particle data collection (EPU) and cryo-electron tomography (TOMO).
  • Imaging Techniques: Features aberration-free image shift (AFIS) and fringe-free imaging (FFI) for increased speed (smaller beam size) and data quality.
  • Imaging Technology: Utilizes the Falcon 4i direct electron detector (320 frames per second) with storage of the data in Electron Event Representation (EER). Live mode enabled using the software Velox.

Applications:

  • Structural Biology – Single Particle Analysis: High-resolution 3D reconstruction of small, complex molecules.
  • Cryo-ET: Limited to simple Cryo-ET experiments – Microscope is not equipped with Energy Filter.

Performance of Krios #2:

Achieves sub-2 Å resolutions, 1.09 Å for Apoferritin – published work in Nature Communications.
